ABSTRACT

Teaching quality is directly related to the performance of universities in fostering talents. Being innovative, high-level, and challenging (IHC) is the basic goal of course reform at universities in the new era. It is essential to reform the contents and teaching mode to improve the IHC properties of the existing courses. We first designed the three-dimensional goals of Molecular Biology Experiment teaching and the contents to support these goals. Then, we pinpointed the common points shared by blended teaching and experiment course, and designed the ways of blended teaching for the course. The reformed course contents and teaching mode have enhanced its IHC properties, and achieved good teaching performance. This paper provides a reference for the reform of experiment courses in universities.

ABSTRACT

OBJECTIVE To study the triterpenoid saponins from Anemone rivularis var. flore-minore and their antitumor activities. METHODS The n-butanol extract of 70% ethanol extract from rhizome of the plant was separated. The triterpenoid saponins were separated and purified by normal silica gel column chromatography ,reversed phase ODS column chromatography , Sephadex LH- 20 gel column chromatography and semi-preparation high performance liquid chromatography. The structures of these saponins were identified by spectral analysis (NMR and MS )and physical and chemical properties. MTT assay was used to test the proliferation inhibitory activity of the compounds against five kinds of human tumor cells (HL-60 cells,A549 cells,HepG2 cells,HeLa cells and U 87MG cells ). The apoptosis inducing effect of compound 7 on U 87MG cells was evaluated by flow cytometric Annexin V-FITC/PI staining test. RESULTS:Sixteen triterpenoid saponins were obtained and identified as 3 β-O-β-D-xylopyranosyl-(1→2)-α-L-arabinopyranosyl-oleanolic acid-28-O-α-L-rhamnopyranosyl- (1→4) -β-D-glucopyranosyl-(1→6)-β-D-glucopyranoside(1),3β-O-L-arabinopyranosyl oleanolic acid- 28-O-β-D-glucopyranoside(2),saponin B (3), 163.com oleanolic acid- 3β-O-β-D-glucopyranosyl-(1→2)-α-L-arabino- pyranoside(4),HN-saponin F (5),clematoside S (6),prosapogenin CP 4(7),cussonside B (8),pulsatilla saponin C (9), clemastanoside D (10),3 β-O-β-D-glucopyranosyl-(1→2)-β-L-arabinopyranosyl-hederagenin-28-O-β-D-glucopyranoside(11), ciwujianoside C 3(12),ciwujianoside A 1(13),huzhangoside D (14),kalopanaxsaponin B (15)and hederacolchiside E (16). Compounds 3,4,6-9 displayed inhibitory activities on the proliferation of tumor cells to different extent ,and compound 7 had the strongest activity ;compound 7 induced the apoptosis of U 87MG cell so as to inhibit the proliferation of cancer cells in a time-dependent manner. CONCLUSIONS The obtained 16 saponins are all identified as oleanolane-type ,among which compound 1 is a new compound. The monodesmosidic saponins ,the sugar chain of which attached at C- 3 and a free carboxyl at C- 28, possess stronger antitumor activity than others.

ABSTRACT

Microbial Engineering is a practical and applied course, it is therefore an indispensable part of teaching to let the students practice in the production line of an enterprise. However, lack of internship resources has always been a bottleneck hampering the high-level internship training during teaching the Microbial Engineering course. A virtual production line developed based on information technology enables students to learn the theoretical knowledge of the complex production system. Moreover, it enables students to perform complex virtual operations, thereby improving students' practicing and innovation abilities. In order to let the virtual simulation experiments project play a great role in undergraduate teaching, this article summarizes the progress, characteristics, and value of the virtual simulation experiments for Microbial Engineering. In addition, the main challenges that limit its shared application are also discussed, with potential solutions prospected.

ABSTRACT

Objective To understand the prevalence rates of HIV-syphilis and HIV-herpes simplex virus 2 (HSV-2) co-infections and related factors among men having sex with men (MSM)who had visited the voluntary HIV counseling and testing (VCT) clinics in Shanghai,China.Methods 756 eligible MSM who attended the VCT clinics of Shanghai Municipality and Putuo district during March to August,2015 were recruited to participate in a cross-sectional survey with questionnaire interview and blood testing for HIV,syphilis and HSV-2.Results A total of 732 participants completed a valid questionnaire survey.The prevalence rates were 3.3% (24/732) for HIV/Syphilis co-infection,1.9% (14/732) for HIV/HSV-2 co-infection,and 0.7% (5/732) for HIWSyphilis/HSV-2 co-infection,respectively.HIV prevalence appeared significantly higher among syphilis-infected participants (45.3%,24/53) than those without Syphilis (7.2%,61/679) (x2=63.11,P<0.001),and was also significantly higher among HSV-2 infected participants (34.1%,14/41) than those without the HSV-2 infection (10.3 %,71/691) (x2 =21.49,P < 0.001).Results from the Multivariate regression analysis indicated that participants who were migrants (0R=3.50,95%CI:1.01-12.17),having had middle school or lower levels of education (OR=4.46,95%CI:1.54-12.87) or ever used illicit drugs (OR=4.25,95% CI:1.67-10.82,P=0.002) were under possible risks on HIV and Syphilis co-infection.Those participants who had high middle school or lower levels of education (OR=6.87,95%CI:1.86-25.42;OR=9.82,95%CI:2.25-42.85) were under risk on HIV and HSV-2 co-infection.Conclusion HIV/Syphilis and HIV/HSV-2 co-infection were seen among MSM who attended the VCT clinics in Shanghai that called for special attention,especially on migrants,those with low education or illicit drug users.

ABSTRACT

Objective To examine the difference in the effectiveness between proximal femoral nail anti-rotation (PFNA) and femoral head replacement (FHR) of intertrochanteric fracture in elderly patients.Methods This study collected 69 patients that underwent PFNA or FHR after intertrochanteric fractures between June 2012 and September 2014.PFNA group was composed of 41 subjects (17 males and 24 females) aged 80-101 years (mean,86.7 years),and time from injury to operation was (3.12 ± 0.95)days.FHR group was composed of 28 subjects (13 males and 15 females) aged 80-99 years (mean,87.2 years),and time from injury to operation was (3.2 ± 1.0) days.Comparison of operation time,intraoperative blood loss,postoperative blood transfusion,off-bed time,complication incidence,hemoglobin concentration and hip function was made between groups.Results Duration of follow-up was 1-3 years (mean,1.8 years).Operation time and intraoperative blood loss in PFNA group were (46.8 ± 4.4) minutes and (77.0 ± 24.2) ml respectively,differed from (57.4 ± 5.9) minutes and (150.7 ± 46.5) ml in FHR group (P < 0.05).Three days after operation,a similar decrease in hemoglobin concentration was seen in PFNA group [(21.3 ±6.1)g/L] and FHR group [(23.2 ±5.8)g/L] (P > 0.05).Off-bed time in FHR group was (3.2 ± 1.2) days,shorter than (7.1-± 2.5) days in PFNA group (P < 0.05).Incidence of postoperative complications in FHR group was 14%,lower than 37% in PFNA group (P < 0.05).Hip function of the two groups presented no significant difference six months after operation,but the proportion of excellent results in FHR group was higher than that in PFNA group (82% vs.73%) (P > 0.05).Conclusion FHR provides better results over PFNA regarding off-bed time,complication incidence and hip function,and hence is preferred for the treatment of elderly patients with intertrochanteric fractures,especially in those with severe osteoporosis.

ABSTRACT

<p><b>OBJECTIVE</b>To screen effective constituents of Discocleidion rufescens against bacteria.</p><p><b>METHOD</b>Compounds were isolated by bioassay-guided fractionation method with various chromatographic methods and their structures were determined by spectral analysis and chemical evidence.</p><p><b>RESULT</b>Seven compounds were isolated from the Et2O and n-BuOH extracts and identified as 5,7-dihydroxy-4'-methoxy flavone (1), quercetin (2), apigenin-7-O-beta-D-glucopyranoside (3), apigenin-7-O-neohesperidoside (4), luteolin-7-O-neohesperidoside (5), gallic acid (6) and beta-daucosteol (7).</p><p><b>CONCLUSION</b>Compounds 1-6 were isolated from genus Discocleidion for the first time and exhibited certain inhibitory activity against Escherichia coli. Compound 6 also showed antibacterial activity against Staphylococus aureus.</p>

ABSTRACT

OBJECTIVE:To explore the protective effect of galactoside-modified oleanolic acid solid lipid nanoparticles(OA-G10SLN)on CCl4-induced acute hepatic injury of rats.METHODS:A total of 60 mice were assigned to normal control group,model group,OA regular solution group or OA-G10SLN group(25.0,12.5,and 2.5 mg?kg-1).Each group was injected from venal caudalis with corresponding drug q.d.for 7d.Acute hepatic injury model was induced by CCl4 on day 6 after medication except for the normal control group.The mice were sacrificed at d7 with serum levels of AST and ALT determined and histopathologic test of liver tissues performed.RESULTS:In OA-G10SLN group compared with model group,the serum levels of AST and ALT decreased markedly(P

ABSTRACT

Objective To investigate the chemical constituents of the sea cucumber Holothuria impatiens collected from the South China Sea.Methods The pure compounds were isolated by extraction,partition and multi-chromatography.Their structures were elucidated on the basis of spectral analysis and chemical evidence.Results Three triterpene glycosides were isolated and identified as holothurin A1(Ⅰ),holothurin A(Ⅱ) and pervicoside C(Ⅲ),together with a physiological substance of adenosine(Ⅳ).Conclusion All were isolated from Holothuria impatiens for the first time.Three glycosides showed significant cytotoxic activities.

Conclusion Laboratory results thus suggest that infection intensity is related to snail size per se, whereas prevalence in the field is related to snail size only through the correlation between size and age. In addition, under these experimental conditions, infected snails were no larger than uninfected snails, so the gigantism snails observed in the field might not be attributable to Trichobilharzia-induced effect.

ABSTRACT

Objective:To screen for the bioactive marine organisms and to study the active constituents from them.Methods:Bioactivities of marine organisms extracts were screened by Pyricularia oryzae bioassay method and the constituents were isolated and identified by a combination of multi chromatography and spectral analysis.Results:Twenty ethanolic extracts and 5 aqueous extracts of the tested marine organisms showed activities causing morphological abnormality of P.oryzae mycelia.Fifty six compounds were obtained and identified from Sargassum carpophyllum,Ishige okamurai and Natarchus leachii freeri ,among which 28 compounds showed activities against P.oryzae and exhibited cytotoxicities on various cultured tumor cell lines,eight compounds showed inhibition effects on Fonsecaea pedrosoi in vitro .Conclusion:The screening data and bioactivity evaluation suggest the P.oryzae bioassay is suitable for preliminary screening of bioactive agents from marine organisms.
